Continuous monitoring of acetonitrile for pharmaceuticals

       Provides continuous monitoring of acetonitrile (ACN): aqueous mobile phase gradients used in reverse phase preparative chromatographic separation of active pharmaceutical ingredients using a ClearView db fiber optic filter photometer.
        Use the spectrophotometer to measure the NIR spectrum of the ACN:water mixture, select the wavelength for the Process Insights process photometer. Samples were measured in 10 mm quartz cuvettes at 40 and 50°C using a 2 m low OH fiber optic cable.
        On fig. 1 shows the obtained spectra for 10, 20, 30, 40 and 50 mass parts. % water in acetonitrile. The main water peak around 1420 nm is outside the chosen optical path length of 10 mm. Another option is to use a 1 or 2 mm optical path to keep this peak at scale. The curves are slightly different for each concentration, showing the effect of sample temperature on the water spectrum.
        Choose two wavelengths for calibration. The calibration model also includes a coefficient for the sample temperature. On fig. 2 shows a graph of predicted and actual results, and shows an excellent calibration with R2 of 0.9997 and standard error of 0.3% water. The temperature coefficients are a correction of 0.15% water/°C. Thus, ClearView db provides excellent accuracy with long-term drift less than 500 µA.
        Process Insights’ low-cost ClearView db photometer simplifies this measurement by using a flow cell that connects directly to the fiber optic probe or bypasses the flow. The ClearView db photometer from Process Insights can measure two independent fiber optic sensors simultaneously, minimizing the total cost of the sampling point.
